Conservation of common ash (Fraxinus excelsior by establishment of a seed orchard with clones with resistance against ash dieback;
Sub-project 2: Phytopathological and gentical characterisation and
vegetative propagation of healthy plus trees
Ash dieback caused by Hymenoscyphus fraxineus) has become to a serious threat for the survival of common ash (Fraxinus excelsior L.) in Europe. The disease proceeds steadily also in
Mecklenburg-Vorpommern. Especially the economical and ecological damages are serious in the coastal region of Mecklenburg-Vorpommern, where ash is growing as sole tree species at wet sites. Therefore planting of ash is essential in the frame of close to nature forestry in Mecklenburg-Vorpommern.
Propagation material with high resistance (determined genetically) against the fungus is needed in sufficient amount for reforestation of damaged sites.
In sub-project 2 plus trees selected in sub-project 1 will be characterised phytopathologically and molecular genetically. These information are necessary for the final selection of the trees used for the seed orchard. Furthermore results were expected to genetical varibility and diversity of ash in the northern part of
Molecular markers will be developed and used for the detection of the fungus in plant material. So it will be possible to test trees which seem to be healthy visually if they are infested by the fungus. Furthermore results are expected concerning the gentical diversity of Hymenoscyphus.
Methods will be improved for vegetative propagation of ash. Selected plus trees resistant to ash dieback will be propagated vegetatively for the planned seed orchard
1. Development and adaptation of methods for the pytopatholgical and molecular genetical characterisation of ash
2. Identification by genotyping of single trees, investigation of diversity and genflow within and between stands of common ash, and comparison of genetic structure of infested and non-infested trees.
3. Characterisation of resistance status of selected plus trees.
4. Vegetative propagation of selected plus trees with high resistance for the establishment of a seed orchard.
In spring 2019 ramets of 60 clones of ash plus trees were planted for the new seed orchard in Mecklenburg-Vorpommern. These clones had shown a high level of resistance in a two-step test before.
